A microplate enzyme-linked immunosorbent assay (ELISA) for detecting antibodies to avian leukosis virus (ALV) of subgroups A and B in infected chickens was developed with the use of Rous-associated virus (RAV)-1 (subgroup A) and RAV-2 (subgroup B) antigens purified by sucrose-gradient centrifugation. The antigen was used for ELISA after treatment with Triton X-100. In the ELISA, the subgroup viral antigen reacted strongly with homologous antiserum but also reacted with heterologous antiserum. Tests with serum absorbed with purified homologous and heterologous virus and tests for antigen-blocking by group-specific antibodies to ALV revealed that the reaction was caused mainly by subgroup-specific antibodies. The ELISA was 8 to 32 times more sensitive than the virus-neutralization (VN) test and detected antibodies to ALV earlier than the VN test in chickens infected experimentally with RAV-1 and RAV-2. In field application of the ELISA, 44.2% of 484 chicken sera were positive for RAV-1 and/or RAV-2 antigen, and 80.4% of flocks were positive. These findings indicate that ELISA is superior to the VN test in sensitivity, simplicity, rapidity, and applicability for large-scale field surveys for ALV infection. 相似文献

Lignan production in callus and cell suspension cultures ofDaphne odora is reported for the first time. Secoisolariciresinol dehydrogenase activity was detected for the first time from Daphne odora and Daphne genkwa (Thymelaeaceae), which are known to produce optically pure (+)-matairesinol. Plesiomonas shigelloides is a causal agent of gastroenteritis, sepsis
and meningitis in humans. We examined the prevalence of P. shigelloides
among great cormorants (Phalacrocorax carbo hanedae) in Japan and the
antimicrobial susceptibility of isolates. P. shigelloides was isolated
from 33 (47.8%) of 69 fecal samples from great cormorants in 2014. All 33 isolates were
subjected to antimicrobial susceptibility testing using broth microdilution methods, which
showed resistance to ampicillin (31 isolates, 93.9%), tetracycline (two isolates, 6.1%)
and trimethoprim (one isolate, 3.0%). The high prevalence of P.
shigelloides in the great cormorants implicates the possible microbiological

Several phenolic compounds were subjected to oxygen-alkali oxidations under oxygen delignification conditions, and their degradations were examined in detail by applying a novel formula. The formula was established on the basis of the two following considerations. The degradation of the phenolic compounds should be expressed by the sum of two types, each of which is caused by molecular oxygen and by highly reactive active oxygen species (HAOS). The degradation should be described by a mathematical equation to which a rate function, k(t), dependent on reaction time t, is applied instead of a rate constant. By rearrangements, the following formula was obtained: k(t) = A/(t + B) (A, B are constants). This is hyperbola, and the illustration of k(t) visualized the contribution of HAOS to the degradation of the phenolic compounds. HAOS did not contribute so much to the degradation, especially at 70 degrees C, which suggests the low energy supply for the HAOS generation at 70 degrees C. The extrapolation of k(t) to the beginning of the reaction gives its initial value, k(initial), which is the rate constant of the reaction between the phenolic compounds and molecular oxygen. As expected, k(initial) was dependent on the electronic property of their substituents. Quantification of the phenolic compounds degraded by HAOS showed that the contribution of HAOS to the degradation is not great. The maximum contribution was observed in the oxidation of 2,6-dimethylphenol at 85 degrees C. In this case, 5 and 95% of the compound were degraded by HAOS and molecular oxygen, respectively. 相似文献

The dependence of the acidolysis reaction of a C6-C3 dimeric nonphenolic β-O-4 type lignin model compound, 2-(2-methoxyphenoxy)-1-(3,4-dimethoxyphenyl) propane-1,3-diol (veratrylglycerol-β-guaiacyl ether, VG), on the type of acid applied was examined using three different acids [0.2 mol/l HCl, 0.2 mol/l HBr,
and 0.1 mol/l (0.2 N) H2SO4 in 82% aqueous 1,4-dioxane at 85°C]. In the HCl system, the major reaction modes of the corresponding benzyl cation-type
intermediate (BC), which is produced by protonation of the α-hydroxyl group of VG and successive release of the water molecule, are the abstraction of the β-proton and hydride transfer from the β-to the α-position. The liberation of formaldehyde from the γ-hydroxymethyl group of BC is the predominant reaction mode in the H2SO4 system. Apparently, an unknown reaction mode or modes is operative in the early stage of the HBr system that causes rapid
disappearance of VG accompanied by the quantitative formation of 2-methoxyphenol without affording the common counterpart
of a Hibbert’s ketone, 1-hydroxy-3-(3,4-dimethoxyphenyl) propan-2-one. The reaction mode in the HBr system changes with the

Magnetic measurements have shown that nondiamagnetic minerals in a lunar crystalline rock of type B are (free Fe(2)+ in paramagnetic pyroxenes) : (antiferromagnetic FeSiO(3)) : (antiferromagnetic FeTiO(3)) : (ferromagnetic iron) = 4.3 : 7 : 20 : 0.08 in weight percentage. The abundance of ferromagnetic Fe in the lunar fines is about 7.5 times its abundance in the crystalline rock. The effects of blending rice flour with wheat flour on bread texture and staling were investigated with three rice varieties with different amylose contents. For the texture analysis of bread crumb, the compression test, puncture test, and tensile test were performed. A flour blend containing rice flour suppressed the recovery of the crumb after compression. For the puncture test, blends with rice flour increased the distance to penetrate the crumb. The rupture strain measured with the puncture test decreased with staling of the bread crumb for all samples tested. The fresh bread crumb sample containing waxy rice flour had much greater extensibility in the tensile test than the other samples tested, but it was dramatically decreased after one day of storage. Endothermic enthalpies corresponding to retrograded amylopectin, which is part of the staling process, were also measured. The enthalpy of bread crumb from the blended flour was lower than that of wheat flour bread up to three days but was higher on day 4. A blend of rice flour thus reduced amylopectin retrogradation during early storage, but it was accelerated later. Bread blended with waxy rice flour showed the lowest enthalpy during storage. 相似文献
